Trypanosoma cruzi Affects Rhodnius prolixus Lipid Metabolism During Acute Infection

چکیده

The interaction between Rhodnius prolixus and Trypanosoma cruzi has huge medical importance because it responds to the transmission of Chagas disease, a neglected tropical disease that affects about eight million people worldwide. It is known trypanosomatid pathogens depend on active lipid endocytosis from insect host meet growth differentiation requirements. However, until now, knowledge how parasite physiology individual organs was largely unknown. Herein, biochemical molecular dynamics triatomine R. metabolism in response T. acute infection were investigated. A qRT-PCR approach used determine expression profile 12 protein-coding genes involved physiology. In addition, microscopic assays revealed droplet levels different identified classes. Finally, spectrometry analyses fatty acid sterol composition their modulation towards infection. downregulated transcript for biosynthetic degrading pathways organs. On other hand, upregulation receptor transcripts indicates an attempt capture more lipids hemolymphatic lipoproteins. Consequently, several classes (such as monoacylglycerol, diacylglycerol, triacylglycerol, cholesteryl ester, phosphatidylcholine, phosphatidylethanolamine) challenge, although modulating only fat body. never leaves gut yet modulates non-infected tissues, suggesting association vector reached by cell signaling molecules. This hypothesis raises intriguing issues inspire future studies parasite-vector field.

Cruzipain Promotes Trypanosoma cruzi Adhesion to Rhodnius prolixus Midgut

BACKGROUND Trypanosoma cruzi is the etiological agent of Chagas' disease. Cysteine peptidases are relevant to several aspects of the T. cruzi life cycle and are implicated in parasite-mammalian host relationships. However, little is known about the factors that contribute to the parasite-insect host interaction. METHODOLOGY/PRINCIPAL FINDINGS Here, we have investigated whether cruzipain could...

Trypanosoma cruzi Infection and Host Lipid Metabolism

Trypanosoma cruzi is the causative agent of Chagas disease. Approximately 8 million people are thought to be affected worldwide. Several players in host lipid metabolism have been implicated in T. cruzi-host interactions in recent research, including macrophages, adipocytes, low density lipoprotein (LDL), low density lipoprotein receptor (LDLR), and high density lipoprotein (HDL). All of these ...

MDL28170, a Calpain Inhibitor, Affects Trypanosoma cruzi Metacyclogenesis, Ultrastructure and Attachment to Rhodnius prolixus Midgut

BACKGROUND Trypanosoma cruzi is the etiological agent of Chagas' disease. During the parasite life cycle, many molecules are involved in the differentiation process and infectivity. Peptidases are relevant for crucial steps of T. cruzi life cycle; as such, it is conceivable that they may participate in the metacyclogenesis and interaction with the invertebrate host. METHODOLOGY/PRINCIPAL FIND...

Effects of Infection by Trypanosoma cruzi and Trypanosoma rangeli on the Reproductive Performance of the Vector Rhodnius prolixus

The insect Rhodnius prolixus is responsible for the transmission of Trypanosoma cruzi, which is the etiological agent of Chagas disease in areas of Central and South America. Besides this, it can be infected by other trypanosomes such as Trypanosoma rangeli. The effects of these parasites on vectors are poorly understood and are often controversial so here we focussed on possible negative effec...

Metalloproteases in Trypanosoma rangeli-infected Rhodnius prolixus.

Protease activities in the haemolymph and fat body in a bloodsucking insect, Rhodnius prolixus, infected with Trypanosoma rangeli, were investigated. After SDS-polyacrylamide gel electrophoresis containing gelatin as substrate, analysis of zymograms performed on samples of different tissues of controls and insects inoculated or orally infected with short or long epimastigotes of T. rangeli, dem...
